I know you guys are to be practical and whatnot but if he calls her "little sister", where's the logic in confessing?
Its a big risk. If you're willing to take it, go for it.
I was in the same boat back in my Jr. Year of highschool. I actually told him and I got "Um.. Sorry I dont feel the same" It took forever for me to build that friendship back. A whole year. But now we're even better friends than before! He's about the sweetest non-real brother ever.

Their friendship is risked.
You just gotta make the choice. If you want him you gotta go out on a limb. Grab him and kiss him if you have to. Maybe he just needs a shock to realize how he really feels about you.
EDIT: some people will bury their feelings because they think they have no chance with that person. His "little sister" act might be because he thinks you are unattainable. But I don't know the guy, so I can't say.
