The story revolves around the title character, Karin (In American commonly known as Chibi Vampire), the middle child in a family of vampires who immigrated to Japan two centuries earlier. However, Karin is different from the rest of the family in that she produces blood instead of feeding on it; because of this, she has to inject blood by biting someone or else she suffers exaggerated nosebleeds. (In manga and anime nosebleeds are also a metaphor for sexual attraction; as Karin is a coming of age story dealing with her first love, the nosebleed imagery does double-duty.) Because of her unusual condition, Karin lives her life just as an ordinary teenage girl would, despite being considered something of a freak by her family. She is able to venture outside during the day and attend high school, and follows the sleep pattern of normal humans. However, the general peacefulness of her life is disrupted with the arrival of a new transfer student, Kenta Usui.

In Karin, vampires (kyūketsuki) are somewhat like traditional vampires of folklore and horror films, however there are also significant differences. Like traditional vampires of folklore they burn up in the sunlight, can control familiars, as well as possessing powers to pin humans down and erase their memories. However, young vampires whose blood-sucking instincts have not yet manifested can withstand subdued daylight and eat human food, but this changes when they hit a certain stage in their development, the vampire equivalent of puberty. From this point on, they become heliophobic and lose their sense of taste for human foods; Ren Maaka described the lunch Karin made for him as tasting like sand.
Karin differs from much vampire literature in that it depicts vampires as not being evil as such, and in fact generally not being interested in humans other than as a source of nutrition.[5] Rather, they are shown almost as victims, their main concern being to remain unnoticed so as to avoid being persecuted (and executed) by humans.
Another difference is that each vampire has his or her own taste in blood based on the personality trait of the human beings on whom they feed. When they suck that person's blood, the associated personality trait is also drained. For example, Karin's taste is unhappiness (不幸, f**ō?), so she is drawn to unhappy people. On biting such a person, that person will lose all their sorrow and become happy, although the effect wears off over time. So in Karin, vampires are able to have a positive effect on humans, but this effect is not intentional[6] and depends on the vampire having a taste for a negative trait. However where their taste is for something positive such as love (as is the case with Karin's grandmother, Elda), the effect of the vampire on human society can be devastating. Since humans and vampires are two completely different species, being bitten by a vampire does not cause a human to become a vampire.
In Karin, the countermeasures against vampires mentioned in vampire mythology are depicted as mostly ineffectual, little more than urban legends. For instance, Karin states that while garlic possesses no magical powers against vampires, it is a combination of the vampires' superhumanly acute sense of smell and the garlic's odor that makes vampires shun it. Similarly, a wooden stake through the heart is not necessarily fatal - both Elda and Henri Marker have survived such - as well as being shot (Ren), stabbed (Elda), run through (Elda and Carrera), skull broken by pistol-whipping (Anju), or even severe burning from prolonged exposure to sunlight (Henri). Their bodies enter a death-like state consistent with the nature of their "fatal" injuries, in which they regenerate completely with no visible scarring, although they will be rather hungry afterwards. Nevertheless, in the manga, vampires as a species are dying out. Those who escaped from the mainland have a low birth rate (zero in the year of the story) while several passed away; the Marker family are the only vampires to have produced children in a century.
In terms of appearance, apart from a few distinguishing features vampires appear to look like normal humans (manga/anime hair and eye color nonwithstanding). The first and foremost non-human feature would be their fangs; under normal situations they appear as nothing more than pronounced canine teeth, but when hungry (or angry) they extend to carotid artery penetration length. Their eyes also appear normal until they are excited, when their round pupils turn into cat-like vertical slits. They also have claw-like fingernails.
Another noticeable feature is their low body temperature due their being nocturnal. As part of their heliophobia, along with their eyes having excellent vision at night yet sensitive to light (both natural and artificial), their bodies have evolved to survive cooler nighttime temperatures. As a result, they tend to overheat if they are out during the day and are at risk of severe sunburn, which could lead to actual burning. In episode 16 of the anime, vampires are executed by prolonged exposure to sunlight, incinerating their bodies into charcoal and ash.
